%20state%20affinity%20constants%2C%20suggesting%20preferred%20dimer%20formation%20as%20MR-MR%5Cu2009%3E%5Cu2009GR-MR%5Cu2009~%5Cu2009GR-GR%3B%20however%2C%20kinetic%20studies%20demonstrated%20that%20GR-containing%20dimers%20had%20the%20longest%20lifetimes.%20Despite%20GR-binding%20differences%2C%20RNA-seq%20identified%20largely%20similar%20subsets%20of%20differentially%20expressed%20genes%20in%20both%20genotypes%20upon%20Dex%20treatment%20%283%5Cu00a0h%29.%20However%2C%20time-course%20experiments%20showed%20gene-dependent%20differences%20in%20the%20magnitude%20of%20expression%2C%20which%20correlated%20with%20earlier%20and%20more%20pronounced%20GR%20binding%20to%20GRE%20sites%20unique%20to%20CO%20including%20near%20Nr3c1.%20Our%20data%20show%20that%20endogenous%20MR%20has%20an%20impact%20on%20the%20kinetics%20and%20differential%20genomic%20binding%20of%20GR%2C%20affecting%20the%20time-course%2C%20specificity%2C%20and%20magnitude%20of%20GC%20transcriptional%20responses%20in%20keratinocytes.%22%2C%22date%22%3A%222023-01%22%2C%22language%22%3A%22eng%22%2C%22DOI%22%3A%2210.1096%5C%2Ffj.202201199RR%22%2C%22ISSN%22%3A%221530-6860%22%2C%22url%22%3A%22%22%2C%22collections%22%3A%5B%2252WELYFA%22%5D%2C%22dateModified%22%3A%222023-01-25T16%3A59%3A21Z%22%7D%7D%5D%7D

key%20regulator%20for%20blast%20survival.%20In%20this%20context%2C%20we%20demonstrate%20that%20contact%20with%20mesenchymal%20stromal%20cells%20promotes%20a%20better%20survival%20of%20blasts%20in%20culture%20in%20the%20presence%20of%20anthracycline%20through%20the%20activation%20of%20ABC%20transporters.%20Stroma-dependent%20ABC%20transporter%20activation%20leads%20to%20the%20induction%20of%20a%20Side%20Population%20%28SP%29%20phenotype%20in%20a%20subpopulation%20of%20primary%20leukemia%20blasts%20through%20alpha%20%28%5Cu03b1%294%20engagement.%20The%20stroma-promoting%20effect%20is%20reversible%20and%20is%20observed%20with%20stromal%20cells%20isolated%20from%20either%20healthy%20donors%20or%20leukemia%20patients.%20Blasts%20expressing%20an%20SP%20phenotype%20are%20mostly%20quiescent%20and%20are%20chemoresistant%20in%20vitro%20and%20in%20vivo%20in%20patient-derived%20xenograft%20mouse%20models.%20At%20the%20transcriptomic%20level%2C%20blasts%20from%20the%20SP%20are%20specifically%20enriched%20in%20the%20drug%20metabolism%20program.%20This%20detoxification%20signature%20engaged%20in%20contact%20with%20mesenchymal%20stromal%20cells%20represents%20promising%20ways%20to%20target%20stroma-induced%20chemoresistance%20of%20AML%20cells.%22%2C%22date%22%3A%222020-04%22%2C%22language%22%3A%22eng%22%2C%22DOI%22%3A%2210.3324%5C%2Fhaematol.2018.214379%22%2C%22ISSN%22%3A%221592-8721%22%2C%22url%22%3A%22%22%2C%22collections%22%3A%5B%2252WELYFA%22%5D%2C%22dateModified%22%3A%222022-03-28T09%3A46%3A23Z%22%7D%7D%5D%7D
